    Image analysis in Gomori´s trichrome stain of skeletal muscles subjected to ischemia and reperfusion injury

    Conditions that produce ischemia and reperfusion injury include orthopedic surgeries, vascular diseases and accidents in remote places in which use of a manual tourniquet is required. Tissues under such stress suffer the consequences of evidenced by changes in their normal microscopic organization that can be reversible or irreversible according to the time and severity of lesion. An experimental model of ischemia has been designed taking into account the characteristics similar to a surgical procedure, from preparation for anesthesia up to the postsurgical follow up of each animal until it finishes the established time of reperfusion. Two muscles, soleus and extensor carpi radialis longus, dissected from Wistar rats that were underwent to short periods of ischemia and short and prolonged periods of reperfusion up to 32 days. There were no significant changes in the macroscopic weight of muscles, but significant differences were found in the area occupied by intramuscular extracellular matrix. During reperfusion, a partial recovery was observed until the last day of study. If we pretend to extrapolate these results to clinical areas, its importance focuses in the recovering of function and the following up of patients after surgical procedures as studied in the present experiment.

    La función tisular se basa en la asociación celular y la comunicación mediante uniones intercelulares o la matriz extracelular, que compone el tejido conectivo. La isquemia conlleva a cambios de lesión a los cuales las células responden según duración e intensidad del estímulo de lesión. En periodos cortos de isquemia y prolongados de reperfusión, el tejido muscular estriado esquelético presenta cambios en la predominancia de los tipos de fibras musculares y en los componentes de la matriz extracelular intramuscular.

    Image analysis of oxidative and glycolytic muscle fibers during reperfusion injury by segmentation based on regions

    Different situations cause ischemia and reperfusion injury, affecting tissues under the level of compression. In this research, abnormal characteristics in distribution of muscle fibers types in soleus and extensor carpi radialis longus, during short periods of ischemia and short and long periods of reperfusion, were determined. Fibers were classified by enzyme histochemistry techniques NADH-TR and myosin-ATPase. Measurements of areas were carried out through semiautomatic image processing by using segmentation based on regions, which evidenced significant changes in distribution during reperfusion followed to one and three hours of ischemia, as well as in comparisons of areas for all periods of reperfusion. This study strengthens the evidence about using practical procedures of image analysis in the diagnosis of tisular abnormalities.
